From Plants with CTAB or High Salt Buffer Protocol

This system contributes to a large on-going research program of the wheat, maize, rape, pea and sunflower genomes. The primary focus is on the genes relevant to disease resistance, agronomic characteristics and crop quality.Genomics contributes to the improvement of varieties used in farming by helping us to understand the functioning of a plant and the genetic diversity of a species. Using genomic information, breeders can now introduce new traits into plants. This can be achieved either by conventional breeding methods using up-to-date knowledge of the genome, or by gene transfer (GMO) if the genetic diversity of a species does not offer possibilities for improvement. Whichever method is chosen, genomics is used to enrich the biodiversity of a species, as well as offer new innovations such as varieties that consume lower quantities of fertilizer, exhibit lower sensitivity to damage from pesticides, exhibit lower sensitivity to climatic hazards, or meet the new quality criteria demanded by the food industry and consumer. HAMILTON's Plant Genomics Workstation was designed for the automation of:

  • DNA extraction

PCR setup Plant gDNA extraction can be performed on the automated system using either a traditional CTAB extraction (for Southern Blot Analysis) or a crude high salt buffer extraction protocol (for PCR Screening). Both extraction protocols automate all the liquid handling, incubation and centrifugation steps, providing a realistic walk-away solution. Using approximately 40mg of frozen ground rape material, the expected DNA yield for the automated CTAB extraction is 80ug. The yield for DNA isolated with the automated crude high salt buffer is approximately 40ug, which can directly be used for PCR set ups.
